NPL and SWISSCI complete a successful Measurement for Recovery (M4R) project

NPL and SWISSCI complete a successful Measurement for Recovery (M4R) project with breakthrough results in crystallisation.

SWISSCI in collaboration with Dr Isabel Moraes, a Principal Research Scientist from NPL, have recently completed a successful M4R project to explore dialysis as an alternative method to established high throughput protein crystallography methodologies.

Using the Diaplate™, a SWISSCI manufactured 96 well high throughput dialysis plate, the NPL science team were able to demonstrate dialysis as a reliable high-throughput method for macro-molecular crystallisation by successfully crystallising 4 different proteins, including 2 membrane proteins.

The membrane proteins crystallised four-fold faster than using any other technique and produced high quality crystals. This data suggests that high-throughput dialysis may have some distinct advantages over other methods and is very simple to employ.

Tristan Kwan of NPL, who performed many of the experiments commented

“Accelerated crystal growth was among some interesting findings and a very welcome surprise. We will employ the technique to expand on the range of crystallisation methods we use in the lab”.

For all the proteins tested, the team were able to grow a high density of microcrystals in certain conditions. Furthermore, the yield of crystals was able to be increased by scaling up the experiment using a higher volume, standalone, SWISSCI manufactured dialysis device, the Diacon™.

 

The results of these experiments indicate that dialysis is an effective method for high throughput protein crystallography and it can be considered as a viable complementary method to the established crystallisation technologies. Initial findings also indicate that it could be a method of choice for production of micro/nano crystals for modern data collection methods including serial crystallography at microfocus beamlines / XFEL’s and MicroED.

The SWISSMX CM supports are designed for high throughput serial data collection from micro-crystals at synchrotrons and XFELS. SWISSMX CMs are designed and engineered by scientists of the Macromolecular Crystallography group and the Laboratory of  Micro and Nanotechnology at the Paul Scherrer Institute in collaboration with the Institute of Polymer Nanotechnology at Fachhochschule Nordwestschweiz.  The polymer-based ultra-thin porous membranes present users with an effective and accessible product for use at MX beamlines.

SWISSCI Plates: Improvements in Imaging with UVXPO

UVXPO: Our Most Advanced Polymer

SWISSCI crystallisation plates are available in 3 polymers.  Polystyrene (PS) is a cost-effective polymer that Is optically clear when using standard bright field or visible light sources. However, when imaging drops using both cross polarised (XP) and UV light, the quality of the image is impaired by the interaction of the polymer with the light source.

SWISSCI’s UVP polymer has been designed to reduce the auto-fluorescence coming from the plate, improving the UV image quality. 

SWISSCI’s UVXPO polymer also has the same low auto-fluorescence while also showing uniform zero background when imaging using cross-polarised light. This makes it our most advanced polymer to date.

UVXPO Sealing Sheets

UVXPO Sealing Sheets

SWISSCI’s UVXPO Sealing sheets are a 50 µm film with 50 µm inert encapsulated silicone adhesive. They are optically clear, with minimal to no auto-fluorescence (UV compatible), and are especially suited for protein crystallization and qPCR applications.

The encapsulated silicone adhesive is non-tacky until pressed against the sealing surface, at which time adhesive is released only in sealing areas to form the strongest available heat-resistant seal around each well on the plate. The adhesive on non-sealing areas of the film, such as directly over sample wells, remains encapsulated and inert.

UVXPO Sealing sheets are sized to fit within the edges of  SWISSCI plates. Two end tabs assist in positioning the film on the plate, and the non-tacky adhesive surface simplifies handling. Easy removal of the end tabs at perforated boundaries prevents lifting and higher evaporation rates that can occur with films that overlap the plate rim.

Cryo EM Grid Box for Transfer and Storage of Vitrified Samples

Grid boxes form an essential part of the Cryo EM workflow as they enable the transfer and storage of vitrified samples on standard EM grids in liquid nitrogen storage dewars. The SWISSCI Cryo EM Grid Box has been specially developed to improve on many features of current solutions. the SWISSCI Cryo EM Grid Box has been developed in close collaboration with the MRC Laboratory of Molecular Biology in Cambridge.

The SWISSCI Cryo EM Grid Box is made from a special polymer formulation for zero static at cryogenic temperatures. The box has four diamond-shaped slot positions with identification numbers. No special tools are required since the lid is rotated with standard laboratory tweezers. The Cryo EM Grid Box is of standard size for all common Cryo EM sample mounting and storage devices.

SWISSCI XPOL LCP for Excellent Crystal Viewing

SWISSCI  XPOL LCP

The new SWISSCI XPol LCP allows excellent crystal viewing for even the smallest of crystals, with uniform zero background when using cross polarised light.

The SWISSCI XPol LCP was developed closely with the Protein Crystallisation Center of the University of Zurich. The center has many years of expertise in crystallizing membrane proteins. The XPol LCP kit has been specially formulated to improve performance versus standard LCP systems.

The Hydrophobic (silanized) Glass Base comes in ANSI/SLAS 1-2004-Standard (SBS) format allowing for easy automation and high throughput screening. The Double-Sided Pressure Adhesive, available in 60μm and 120μm, comes readily stuck to the Glass Base with space for barcode application. The Double-Sided Pressure Adhesive has 96 holes which are 7mm in diameter and have a 2.3μ volume in the 60μm version and 4.6μl volume for 120μm. The large diameter almost eliminates the occurrence of precipitation solution being pulled to the side, which can often happen with smaller diameter holes.

The SWISSCI XPol LCP comes as standard with 175μm Cover Glass. For the addition of UV/Flu, UV Cover Films are also available upon request at 40μm, 100μm or 200μm thickness.

SWISSCI LCP Modular

The SWISSCI LCP Modular range aims to offer more flexibility to the membrane protein crystallographers LCP experiment. There are three choices of LCP Bases which are combined with one of five pre-affixed LCP Double Sided Pressure Adhesives*. The Pressure Adhesives are of varying thickness and well dimensions. To complete the LCP sandwich plate there is a choice between two sizes of Cover Glass or three UVP Cover Films that are of varying thickness.
